Flat tyre!

Really hard to push into the shed, very flat, bad thought when in the car that the screw may actually of come off the bike!
What's the deal? New tyre?

Hard to tell from the pic, but how much tread depth left? Assuming there's plenty of meat left you can get it plugged as long as the hole isn't too big. If it's more than half worn I'd probably cut my losses just get a new tyre. I've had about four or five plugs in the life of my current bike, and thankfully all were fixable. Your local bike shop will tell you if they can fix it or not. Being near the middle is good, the further towards the edge, the smaller the repair is allowed to be.

How do I get it to the bikeshop though? I know they stock Bridgestone but no idea how to get them the bike (I can't push it on my own no wonder ride it!)

take the wheel off the bike and get a lift of a friend or family

As above... Pop the wheel off and take it down to your local tyre shop. From the pictures alone ot looks like you have a good chance of that being plug-able as it is near the middle of the tyre.
No idea what a plug costs nowadays - but it is deffinately cheaper than new rubber
As for if the "screw" came from your bike....there really isn't that many "screws" I can think of on the bike. Possible a few holding the fairing / V piece but thats all I can think off (sure sombody else will think of some more). Check if they are all there...if so I would guess it didn't come from your bike

I cover all my screws on my belly pan with gaffer tape squares so theres no chance of them falling off and landing in my wheel.

Could be off the V piece, I put three new ones in the other day, haven't checked yet.
What tools do I need to get it off?

What size is the rear wheel nut? Haynes shows taking it off by hand and I don't have callipers to measure it's size
